Ordered to be built in 1536, it was part of an extensive artistic program, which included its Palace, a University and a Hospital, destined to heighten the fame, fortune and glory that the personal secretary of Charles V had achieved; for which he turned to top-level artists.
The project was carried out by Andrés de Vandelvira starting in 1540. The temple was consecrated in 1559. Its first chaplain was Dean Ortega, for whom the large palace to the left of the main façade of the chapel was built.
